体内心肌缺血微环境下大鼠骨髓间充质干细胞钾离子通道的表达

Summary:目的动态观察在体内心肌缺血微环境下,大鼠骨髓间充质干细胞(mesenchymalstemcells,MSCs)向心肌细胞分化过程中主要钾离子通道的分化表达。方法采用左冠状动脉前降支(1eftanteriordescendingcoronaryartery,LAD)结扎术建立大鼠急性心肌梗死(myocardialinfarction,MI)模型(n=60),将绿色荧光蛋白(greenfluorescentpro—rein,GFP)标记的MSCs,心外膜下注射移植到心肌梗死周边区;免疫荧光染色观察移植后3d(MI一3d组)、5d(MI-5d组)、7d(MI-7d组)、9d(MI9d组)MSCs的存活情况及其主要钾离子通道(Kvl.4、Kir2.1、Kv4.3、Kcal.1)蛋白的表达;应用激光捕获显微切割技术分离以上各组心肌组织中GFP标记的MSCs群,应用Real—timePCR测定其Kvl.4、Kir2.1、Kv4.3、Kcal.1的mRNA表达。结果免疫荧光染色观察到移植的MSCs于移植后第3天开始持续表达Kvl.4、Kir2.1、Kv4.3,不表达Kcal.1,于移植后第9天几乎未观察到MSCs;Real—timePCR结果显示移植后3、5、7d,移植的MSCs上Kvl.4和Kir2.1表达呈逐步增多趋势,差异具有统计学意义(P〈0.05),Kv4.3的表达无此变化趋势,但高于未移植MSCs,未见明显差异。结论移植的MSCs在大鼠体内心肌缺血微环境下向心肌细胞分化的过程中能够表达部分钾离子通道表型,但其不能长时间、有效的存活。
