抑制人肝星状细胞株迁移的中药组分筛选

目的从6种已证实有抗肝纤维化作用的中药组分中,筛选出抑制肝星状细胞迁移的最佳者。方法培养人肝星状细胞株(LX-2),用细胞迁移杯培养方法分别观察不同浓度梯度(10-4、10-5、10-6、10-7mol/L)的青蒿琥酯、灯盏花素、红景天苷、丹参酚酸B盐、粉防己碱、黄芪总甙对血小板衍生生长因子-BB(PDGF-BB)(10 ng/ml)诱导的LX-2迁移的抑制,从而筛选出各组分最佳作用浓度;随后比较各组分最佳浓度对LX-2迁移的抑制作用,而筛选出最佳组分。结果 6种中药组分均可抑制PDGF诱导的LX-2的迁移;但10-4mol/L丹参酚酸B盐、10-4mol/L黄芪总甙、10-4mol/L与10...

Objective To screen the best component from six of components of traditional Chinese medicine which were confirmed with the anti-fibrosis function.Methods A trans-well chamber system was used to observe the changes of the human hepatic stellated cell line(LX-2) migration.We select human HSC cell line(LX-2) which induced by platelet derived growth factor-BB(PDGF-BB) to study the effect of inhibition of migration.Cells were incubated with 10-4mol/L-10-7mol/L concentration of Artesunate,Breviscapine,Salidroside,Salvianolic acid B,Tetrandrine,Astragalosides,respectively.Results Six kinds of Chinese medicine components can inhibit the PDGF-induced migration of LX-2.10-4mol/L Salvianolic acid B,10-4mol/L Astragalosides,10-4mol/L Tetrandrine and10-5mol/L Tetrandrine have different affectiveness on cell morphology.Based on the principles of the selection,10-4mol/L Artesunate,10-4mol/L Breviscapine,10-5m
