广西薄皮甜瓜疫病菌的鉴定

近年来,甜瓜疫病是广西露地栽培甜瓜产区发生最严重的病害之一,为明确引起甜瓜疫病病原的分类地位,对该病原的形态学、致病性及r DNA-ITS序列进行分析研究,结果表明:经分离纯化获得的病原菌,接种后出现的症状与直接采集到的病害标样症状一致,确定其为致病菌,对病原菌进行形态学观察,该病原菌的菌丝无隔,孢子囊形态大小不一,具乳突,经培养镜检,未见有卵孢子产生,将待测菌株r DNA-ITS序列与Gen Bank中相关菌株ITS序列进行同源性比较,结果显示该菌与辣椒疫霉的同源性最高。结合病原菌形态学特征及菌株r DNA-ITS序列,将引起广西薄皮甜瓜疫病的病原菌鉴定为辣椒疫霉(Phytophthora...

Bibliography:Phytophthora blight is one of the most serious threats to production of oriental melon in Guangxi province recently. It usually reduces the quality and yield of oriental melon. In this study,the causal organism of oriental melon blight was identified by pathogenicity,morphology and ribosomal DNA-ITS. The results showed that the isolated fungus was used for infecting healthy oriental melon fruit. Disease symptoms after inoculation were the same as that on the collected samples,which showed the fungus was the pathogen. Hyphae of the pathogen was transparent and aseptate. Sporangia are papillate and variable in shape and size. The oospores were not found after cultivate for 20 days. Comparing the measured sequence with the related sequence of Phytophthora capsici in Gen Bank,the homology was 97 %. Based on its morphological characteristics and r DNA-ITS sequences analysis,this fungus was identified as P. capsici Leonian.
