液化淀粉芽孢杆菌YM3发酵配方研究

为筛选液化淀粉芽孢杆菌(Bacillus amyloliquefaciens)YM3的发酵配方,初期以摇瓶进行培养,确定最适配方并以此配方进行培养条件筛选。结果表明最适配方为糖蜜0.5%、乳糖1%、大豆分离蛋白2%,以此配方进行培养条件研究,接种24 h内菌量显著提升1000倍,24~8 h菌量显着提升10倍,此后培养菌量无显着差异。其他培养条件如不同温度、p H之间菌量则无显着性差异。用10 L发酵槽测试,显示以糖蜜0.5%、乳糖1%、大豆分离蛋白2%作为培养基,转速200至300 r/min、温度30℃、p H无调整,培养5 d,可得到菌量4×109CFU/m L以上之发酵液。利用1 t大...

Bibliography:The aim of this study was to use Bacillus amyloliquefaciens( YM3) to conduct fermentation formula test. [Methods]At the beginning,the shake flask was used to culture for test using shake,in order to find the medium fermentation formula. Then culture condition test was conducted with these formulae. The medium fermentation formula was 0. 5 % molasses,1 % lactose,and 2 % soybean peptone,with which fermentation formula test was conducted. The result showed that number of bacterial increased 1000 times in 24 h,10 times between24- 48 h,and there was no significant differences after 24 h. Other culture conditions such as temperature and p H value showed no significant differences. Through test of 10-liter fermentation tank,YM3 reached〉 4 × 109 CFU / m L by using 0. 5 % molasses,1 % lactose,2 %soybean peptone as the culture medium for five days under the conditions of stirring speed from 200 to 300 r / min,30 ℃,no adjustment of p H value. As for 1-ton fermntation tank using the same culture medium,YM3 reached 〉6 × 1
