芽孢杆菌C产亚硝酸还原酶的发酵培养基优化及酶学性质研究
通过单因素试验和响应面分析对亚硝酸还原酶产生菌c的发酵条件进行优化,获得最佳发酵培养基为:葡萄糖2.26%,蛋白胨0.2%,NH4C10.12%,K2HPO4·H2O0.2%,MgSO40.075%,KCl0.1%,FeSO40.004%,NaNO20.2%,在此条件下,酶活达到317.82U/mL,比优化前的190.26U/mL提高了67.05%。产生的亚硝酸还原酶的酶学性质表明,该酶被Mn^2+、Mg^2+、Na^+激活,受Fe^2+、EDTA抑制;最适温度和pH值分别为30℃和7.4;当温度低于50℃、pH值在7.0~8.0时,酶能保持较高的活性。与现有文献报道的筛选细菌相比较,该菌株亚...

C were optimized by single factor experiment and response surface analysis. The optimal fermentation conditions were as follows: 2.26% glucose, 0.2% peptone, NH4Cl 0.12%, K2HPO4 ·H2O 0.2%, MgSO4 0.075%, KC1 0.1%, FeSO4 0.004%, NaNO2 0.2%.
