广西淮山根腐线虫病病原鉴定

【目的】明确广西淮山根腐线虫病病原种类,为生产上淮山根腐线虫病的防治提供理论依据。【方法】从广西淮山主产区采集有病斑的淮山块茎,连同根际土壤一起带回实验室;通过直接浸泡分离法和改良贝曼漏斗法分离线虫,应用形态学和分子生物学方法对分离获得的线虫进行种类鉴定。【结果】从淮山病薯和病土中分离到一种线虫,其块茎虫口密度最高达704头/cm~2。线虫虫体蠕虫形,较为短粗,唇区低平,口针粗而短基部球宽,圆形;食道腺覆盖肠腹面,中食道球十分显著,卵圆形雌虫阴门横裂,位于虫体后端,确定为短体线虫属。分离的线虫与咖啡短体线虫的28S rDNA序列同源性最高,与两个已知的咖啡短体线虫在系统发育树上形成一个分支,验...

[Objective]The species of nematode causing root rot disease of Chinese yam was identified,in order to controlling root-rot nematode disease of Chinese yam in Guangxi.[Method]The Chinese yam tuber with disease spot and rhizosphere soil were collected from main producing areas of Guangxi.And the nematodes were isolated by immersion and its modified Baermann funnel methods.Then isolated nematodes were identified based on their morphological and molecular biological characteristics.[Result]A species of nematode was isolated from diseased tuber and rhizosphere soil,the highest density of nematode in diseased tuber reached up to 704 nematodes per square centimetre.The morphological characters of nematode were as follows:helminthoid and dumpy body,flat lip,short and thick scalpellum,massive round basal knobs,intestine venter covered by esophageal gland,oval mid-esophagus spheroid,transverse dehiscence of female vulva located in posterior of body.It could be deduced that isolated nematodes were identified as
