猪不同泌乳期母乳外泌体中let-7a和let-7f的差异表达研究

【目的】为研究miRNAs在仔猪免疫系统发育过程中的调控作用。【方法】本研究采用荧光定量PCR技术(Q-PCR)研究了不同泌乳期(0、3、7、14、21和28 d)的梅山猪和大约克夏猪母乳外泌体中let-7a和let-7f的差异表达。【结果】let-7a和let-7f在泌乳早期(0~3 d)的表达丰度明显高于泌乳后期(7~28 d);品种间分析显示梅山猪母乳外泌体中let-7a和let-7f在整个泌乳期(0~28d)的表达水平均明显高于大约克夏猪。【结论】研究表明,let-7a和let-7f能够正向调控仔猪免疫系统发育。本研究可为阐明母乳外泌体包被的miRNA在幼畜免疫系统发育过程中的具体调控...

[ Objective] It is of great interest to understand the mechanism underlying breast milk quality for immune disease research. [ Meth- od ] The expression changes of let-7a and let-Tf miRNAs were analyzed in porcine milk exosomes across the entire lactation period (newborn to 28 days after birth) by quantitative real-time PCR. [ Result ] let-7a and let-Tf were more abundant in the early lactation period (0 and 3days) than in the later lactation period (7, 14, 21 and 28 days), and were expressed at higher levels in Meishan pigs compared with York- shire pigs throughout the entire lactation period. [ Conclusion] This study is a preludetoin-depth investigations of the essential roles of breast milk exosome-packaged miRNAs in the development of the infant' s immune system.
