樱花新品种白玉吉野组培快繁技术研究

以白玉吉野当年生带腋芽茎段为外植体,进行了外植体的灭菌、启动、增殖和生根培养试验。试验结果表明:外植体灭菌消毒方法为75%乙醇浸泡30 s,0.1%HgCl_2浸泡6 min,成活率达51.33%;促进腋芽诱导最佳培养基配方:MS+2.0 mg/L BA+0.2 mg/L IBA,诱导率为72.65%;适宜的增殖最佳培养基配方:MS+2.5 mg/L BA+0.2 mg/L NAA,增殖系数为5.76;诱导生根率最高的培养基配方为MS+0.5 mg/L BA+0.2 mg/L NAA,生根率达92.02%。...

The one-year-old stems with axillary buds of oriental cherry new variety Baiyujiye were selected as explants,and the experiments in the sterilization,initiation,proliferation and rooting culture of the explants were carried out. The results showed that the best sterilization procedure for explants was 0.1% HgCl_2 for 6 minutes after 75% alcohol for 30 seconds,and their survival rate could reach 51.33%. The optimal medium for the inducement of axillary bud was MS+ 2.0 mg/L BA+ 0.2 mg/L IBA,and the inducement rate was 72.65%. The optimum medium for the proliferation was MS+ 2.5 mg/L BA+ 0.2 mg/L NAA,and the proliferation coefficient was 5.76. The best medium for the rooting was MS + 0. 5 mg/L BA + 0. 2 mg/L NAA,and the rooting rate reached 92.02%.
