南美白对虾多酚氧化酶的提取及其性质研究
以南美白对虾为对象,研究了多酚氧化酶(PPO)分离提取工艺及其生化特性。在单因素试验基础上,采用响应面试验优化了料液比、提取液pH值和浸提时间等因素提取南美白对虾PPO的工艺,并对PPO的生化性质进行分析。结果表明:液料比为2∶1、提取液体系pH值为8.0、浸提时间为3 h等条件下,提取的PPO具有最大酶活力为94.8 A/min·mL,所得回归模型显著(P〈0.0001),方程拟合性好(R-2=0.9762),可用于预测南美白对虾中PPO的提取;PPO酶促反应动力学方程符合米氏方程,米氏常数Km为30.322 mmol/L,V(max)为0.281 A/min。PPO性质研究将为南美白对虾黑...

Notes | 44-1267/S The optimization of extraction and biological characteristics of polyphenol oxidase ( PPO ) of white shrimp ( Penaeus vannamei ) were studied. The experimental factors and levels were determined by single factor experiments. Response surface methodology was used to explore the effects of solid-liquid ratio, pH and extraction time on PPO activity. The results showed that the optimal extraction conditions were as follows: the solid-liquid ratio was 2 : 1, pH was 8.0 and the extraction time was 3.0 h. Under the optimal extractions, the PPO activity could reach 94.8 A/rain - mL. Regressive model of extracted PPO was significant ( P 〈 0.0001 ) and better fitting ( R^2=0.9762 ), it could be used to predict the PPO extraction. The kinetic equation of enzyme reaction was consistent with Michaelis Menten equation. The K,, was 30.322 mmol/L and Vmax was about 0.281 A/min.
