HPLC法测定癸氧喹酯壳聚糖-海藻酸钠微囊中药物含量
建立癸氧喹酯壳聚糖-海藻酸钠微囊中癸氧喹酯的HPLC含量测定方法。采用破囊液(O.2mol/LNaHC03和0.06mol/LNa3c6H507·2H20,pH=7.8—8.2)对癸氧喹酯壳聚糖-海藻酸钠微囊进行破囊,破囊后用甲醇和三氨甲烷混合溶液(4:1)超声提取癸氧喹酯。HPLC检测方法采用AgilentZORBAX80AExtend-C18色谱柱;以乙腈-0.1%甲酸水溶液(75:25)为流动相;检测波长为267nm;流速为1mL/min。结果表明,所采取的破囊方法破解效率商,微囊破解完全。方法精密度与准确度表明,癸氧喹酯浓度在0.05-10μg/mL的范围内线性关系良好,回归方程为Y=...
