凡纳滨对虾Peroxinectin基因部分cDNA序列的克隆及表达分析

为研究病毒感染凡纳滨对虾血细胞黏连因子基因表达量的变化,进而研究该因子在对虾先天免疫系统中的作用,利用RACE方法克隆凡纳滨对虾血细胞黏连因子cDNA序列,并通过荧光定量PCR方法分析其在病毒感染前后不同组织中的表达情况。成功获得了长度为1529bp的Pemxinectin基因部分eDNA编码序列,3’端含有一段长度为319bp的非翻译区,推测血细胞黏连因子部分序列编码402个氨基酸。凡纳滨对虾血细胞黏连因子的氨基酸序列高度保守,在线比对发现其氨基酸序列与斑节对虾和中国明对虾相似性达到90%,而与锯缘青蟹有87%的相似性。通过荧光定量PCR技术检测IHHNV感染前后血细胞黏连因子基因表达量的变...

The changes of Litopenaeus vannamei peroxinectin expression after virus infection and the role of peroxinectin in immune system were studied. L/topenaeus vanname/peroxinectin gene was cloned and analyzed using RACE method, and the expression of the gene was analyzed by real-time fluorescence PCR. In this study, a partial cDNA of Litopenaeus vannamei peroxinectin was cloned. The cDNA sequence was 1529bp. The 3' end contained a 319bp-untranslated region and encoded 402 putative amino acids, which exhibited a significant similarity to PX of Penaeus monodon( 90% ), Fenneropenaeus chinensis( 90% ) and Scylla serrata( 87% ).
