RhoA与2型糖尿病大鼠心肌病变的关系及法舒地尔的干预作用

目的探讨RhoA在糖尿病心肌病变中的作用及法舒地尔治疗的影响。方法以高脂高糖饲料联合小剂量链脲佐菌素(30mg/kg)腹腔注射制备2型糖尿病sD大鼠模型(n=30),将成模大鼠随机分为糖尿病组(n=15)和法舒地尔干预组(n=15),正常sD大鼠(n=10)作为对照组。法舒地尔干预组给予法舒地尔10mg·kg-1·d-1腹腔注射,糖尿病组和对照组均给予等体积生理盐水腹腔注射。药物干预持续4周。实验末,取大鼠心脏,称重并计算心脏肥厚指数。取左心室组织,进行光镜、电镜观察;应用免疫组化法和逆转录.聚合酶链反应分别检测RhoA在心肌组织中的蛋白表达(用累积吸光度值表示)及mRNA表达。两组数据比较采...

Summary:目的探讨RhoA在糖尿病心肌病变中的作用及法舒地尔治疗的影响。方法以高脂高糖饲料联合小剂量链脲佐菌素(30mg/kg)腹腔注射制备2型糖尿病sD大鼠模型(n=30),将成模大鼠随机分为糖尿病组(n=15)和法舒地尔干预组(n=15),正常sD大鼠(n=10)作为对照组。法舒地尔干预组给予法舒地尔10mg·kg-1·d-1腹腔注射,糖尿病组和对照组均给予等体积生理盐水腹腔注射。药物干预持续4周。实验末,取大鼠心脏,称重并计算心脏肥厚指数。取左心室组织,进行光镜、电镜观察;应用免疫组化法和逆转录.聚合酶链反应分别检测RhoA在心肌组织中的蛋白表达(用累积吸光度值表示)及mRNA表达。两组数据比较采用t或tw检验。结果糖尿病组大鼠心脏肥厚指数(0.006O±0.0004)高于正常对照组(0.0030±0.0003)(t=21.121,P=0.005),法舒地尔干预组大鼠心脏肥厚指数(0.0050±0.0004)低于糖尿病组(t=4.523,P=0.008)。光镜、电镜观察显示糖尿病组心肌病理改变明显,法舒地尔干预组心肌病理变化减轻。免疫组化分析示糖尿病组大鼠心肌RhoA的累积光密度值(70±10)是对照组(24±3)的2.89倍,法舒地尔干预组(53±6)比糖尿病组降低了25%。糖尿病组大鼠心肌RhoAmRNA表达(1.43±0.16)高于正常对照组(0.42±0.06)(t=-33.437,P=0.012),法舒地尔干预组(0.73±0.13)较糖尿病组减少(t=8.145,P=0.038)。大鼠心肌RhoA的蛋白表达水平及mRNA表达水平与心脏肥厚指数均呈正相关(r=0.94、0.85,均P〈0.05)。结论RhoA与糖尿病心肌病变的发生发展存在密切关系,法舒地尔通过下调RhoA的表达发挥对2型糖尿病心肌的保护作用。

Objective To investigate the association between RhoA and cardiomyopathy in type 2 diabetic rats and the intervention effect of fasudil. Methods The type 2 diabetic SD rat models were established by feeding with high-fat, high-sucrose diet and intraperitoneal injection of low-dose streptozotocin (30 mg/kg). The type 2 diabetic rats (n=30) were randomly divided into two groups: diabetic group (n=lS) and fasudil group (n=lS). In addition, 10 SD rats were set as normal control. The rats in fasudil group received fasudil at a dosage of 10mg·kg-1·d-1 by intraperitoneal injection for 4 weeks. The rats in diabetic group and control group were given injection of saline simultaneously.
