Summary:目的 探讨Sestrin2/一磷酸腺苷活化的蛋白激酶(AMPK)介导的细胞自噬在糖尿病大鼠心肌缺血再灌注损伤中的作用及叔丁基对苯二酚(TBHQ)预处理的保护作用.方法 清洁级健康成年雄性SD大鼠60只,体重200~220 g,腹腔注射1%链脲佐菌素60 mg/kg制备大鼠糖尿病模型,采用随机数字表法分为6组(n=10):正常大鼠对照组(NS组)、正常大鼠缺血再灌注组(NIR组)、正常大鼠TBHQ预处理组(NIPC组)、糖尿病大鼠对照组(DS组)、糖尿病大鼠心肌缺血再灌注组(DIR组)、糖尿病大鼠TBHQ预处理组(DIPC组).结扎冠状动脉左前降支30 min,再灌注120 min建立心肌缺血再灌注模型,NIPC组、DIPC组实验前连续5 d腹腔注射TBHQ 40μg/kg.苏木素伊红染色观察心肌组织,酶联免疫法检测肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)水平,Western blotting测定心肌组织Sestrin2、磷酸化AMPK(p-AMPK)、自噬效应蛋白Beclin1、半胱氨酸天冬氨酸蛋白酶3(Caspase3)的表达.组间比较采用单因素方差分析.结果 与NIR组相比,NIPC组CK-MB、LDH活性降低(343±37比453±36、428±65比576±45,t=0.0066、0.0134,均P〈0.05),SOD活性升高(706±17比566±21,t〈0.0001,P〈0.05),Sestrin2、p-AMPK、Beclin1表达显著升高(t=0.0004~0.0056,均P〈0.05),Caspase3表达降低(t=0.0009,P〈0.05).与NS组相比,NIR组和NIPC组CK-MB、LDH、SOD活性显著升高(F=17.04~413.16,均P〈0.05),与DS组相比较,DIR组和DIPC组CK-MB、LDH、SOD活性显著升高(F=91.83~183.28,均P〈0.05),Sestrin2、p-AMPK、Beclin1、Caspase3表达显著升高(F=8.63~128.69,均P〈0.05).与DIR组相比较,DIPC组CK-MB、LDH活性、Caspase3表达显著降低(t=0.0080~0.0227,均P〈0.05),SOD活性、Sestrin2、p-AMPK、Beclin1表达明显升高(t=0.0003~0.0069,均P〈0.05).结

Objective To investigate Sestrin2/adenosine monophosphate-activated protein kinase (AMPK)induced autophagy in myocardial ischemia-reperfusion injury in diabetic rats and the protective effect of tert-butyl hydroquinone (TBHQ). Methods Healthy adult male SD rats were randomly divided into 6 groups (n=10): control rats with sham surgery group (NS), control rats with ischemia-reperfusion (NIR)group, control rats with ischemia-reperfusion+TBHQ group (NIPC), diabetes rats with sham surgery group (DS), diabetic rats with ischemia-reperfusion(DIR) group, diabetic rats with ischemia-reperfusion+TBHQ group(DIPC).
