消癌平联合顺铂对高转移人卵巢癌HO-8910PM细胞的抑制作用
目的探讨消癌平和顺铂对高转移人卵巢癌HO-8910PM细胞增殖、凋亡和侵袭的影响,及其对裸鼠皮下移植瘤生长的影响。方法将不同浓度消癌平注射液和(或)顺铂作用于体外培养的高转移人卵巢癌HO-8910PM细胞,采用CCK-8法检测细胞的增殖抑制率,流式细胞术检测细胞周期和细胞凋亡,Transwell实验检测细胞的侵袭能力。建立裸鼠皮下HO-8910PM细胞移植瘤模型,比较药物作用后移植瘤体积的差异。结果0.1、0.5、1.0、1.5、2.0mg/ml消癌平作用于HO-8910PM细胞后,细胞增殖受到抑制,随着消癌平浓度的增加,细胞增殖抑制率增高。1.0mg/ml消癌平作用HO-8910PM细胞48...

Summary: | 目的探讨消癌平和顺铂对高转移人卵巢癌HO-8910PM细胞增殖、凋亡和侵袭的影响,及其对裸鼠皮下移植瘤生长的影响。方法将不同浓度消癌平注射液和(或)顺铂作用于体外培养的高转移人卵巢癌HO-8910PM细胞,采用CCK-8法检测细胞的增殖抑制率,流式细胞术检测细胞周期和细胞凋亡,Transwell实验检测细胞的侵袭能力。建立裸鼠皮下HO-8910PM细胞移植瘤模型,比较药物作用后移植瘤体积的差异。结果0.1、0.5、1.0、1.5、2.0mg/ml消癌平作用于HO-8910PM细胞后,细胞增殖受到抑制,随着消癌平浓度的增加,细胞增殖抑制率增高。1.0mg/ml消癌平作用HO-8910PM细胞48h后,细胞增殖抑制率为(38.3±4.5)%;消癌平联合顺铂作用后,细胞增殖抑制率为(53.4±3.0)%,差异有统计学意义(P〈0.05)。消癌平组和顺铂组中G0/G1期细胞的比例分别为(74.1±1.6)%和(68.6±1.6)%,与对照组[(64.2±1.6)%]比较,差异均无统计学意义(均P〉0.05);经消癌平和顺铂联合作用后,G0/G1期细胞比例为(79.9±1.7)%,与顺铂组比较,差异有统计学意义(P〈0.05)。1.0mg/ml消癌平和10.0ug/ml顺铂处理HO-8910PM细胞48h后,早期凋亡细胞的比例分别为(16.1±1.6)%和(35.6±1.6)%,与对照组[(2.2±1.6)%]比较,差异均有统计学意义(均P〈0.05);联合用药组早期细胞凋亡的比例为(59.9±1.8)%,与消癌平组和顺铂组比较,差异均有统计学意义(均P〈0.01)。顺铂和消癌平联合作用HO-8910PM细胞后,侵袭至滤膜下表面的细胞数为(89.2±20.7)个/视野,低于对照组[(187.2±24.6)个/视野]、消癌平组[(141.8±13.7)个/视野]和顺铂组[(155.8±19.4)个/视野],差异均有统计学意义(均P〈0.05)。顺铂组、消癌平组和联合用药组裸鼠的移植瘤抑瘤率分别为34.2%、23-4%和59.0%,联合用药组的抑瘤率高于顺铂组和消癌平组,差异均有统计学意义(均P〈0.01)。结论消癌平能抑制高转移人卵巢癌HO-8910PM细胞增殖,引起G0/G1期细胞阻滞,促进细胞凋亡,降低HO-8910PM细胞的侵袭能力。消癌平联合顺铂可以产生协同抗肿瘤作用。 |

Bibliography: | Objective To explore the effect of a Chinese medicine, Xiaoaiping, in combination with cisplatin on the proliferation, invasion and apoptosis in human ovarian cancer HO-8910PM cells in vitro and vivo. Methods CCK-8 assay was used to detect the inhibitoty effect of Xiaoaiping alone or in combination with cisplatin on the proliferation of human ovarian cancer HO-8910PM cells. Flow eytometry was used to detect the apoptosis and cell cycle distribution. Transwell migration test was used to assay the effect of drugs on the cell invasive capability. Changes of the tumor volume in nude mice were observed to evaluate the antitumor effects in vivo. Results CCK-8 assay Xiaoaiping alone or combined with cisplatin could inhibit the proliferation of HO-8910PM cells with a dose-dependent manner. The inhibition rates of Xiaoaiping combined with cisplatin were (53.4±3.0)%, significantly increased than those with single drug (P〈 0. 05).
