Effects of a 6-O-Hydroxyethyl Group on the Hydrolysis of 6-O-Hydroxyethylchitin (Glycolchitin) by Chitinase

Kinetic experiments were done on a chitinase from Bacillus sp. with four kinds of 6-O-hydroxyethylchitin (HE-chitin, glycolchitin) with different degrees of substitution (ds) for the 6-O-hydroxyethyl group as substrates. The K m increased markedly by increase in the ds, and the value for HE-chitin w...

Summary:Kinetic experiments were done on a chitinase from Bacillus sp. with four kinds of 6-O-hydroxyethylchitin (HE-chitin, glycolchitin) with different degrees of substitution (ds) for the 6-O-hydroxyethyl group as substrates. The K m increased markedly by increase in the ds, and the value for HE-chitin with the ds of 1.0 was 4.5-fold higher than that for a substrate with the ds of 0.33. In contrast, the V max decreased in response to an increase in the ds, and the value for the former substrate was about a half of that for the latter one.
