Summary:An active porcine Follicle-Stimulating Hormone (rFSH) was secreted by a recombinant strain of the methylotrophic yeast Pichia pastoris. The influence of the carbon source, either methanol or a mixture of methanol with glycerol or sorbitol, on rFSH production was subsequently studied. On a sorbitol/methanol medium using a high cell-density fed-batch culture the rFSH concentration was 93 mg l −1. The addition, in fed-batch culture, of vitamins and trace elements to the synthetic medium allows optimal biomass and recombinant protein production to be attained. Under these conditions, 187 mg l −1 of rFSH were produced and the production yield Y rFSH/X reached 2.5 mg rFSH g DCW −1. In continuous culture the carbon source and growth factor supplementation had an identical effect on rFSH synthesis, which reached a maximum concentration of 280 mg l −1 and a production yield of 3.6 mg rFSH g DCW −1.
