Summary: | We studied the changes induced by oncogenic transformation on fibroblast-fibrin interactions
in vitro
. The test system used was retraction of fibrin clots formed by clotting a mixture of cells to be tested and platelet-free plasma with thrombin. Fibroblasts were studied from healthy donors of different ages, from subjects with genetic abnormalities and from various cancer patients. As a reference system, established human cell lines were tested. The ability to interact with fibrin was completely lost in cells with an infinite replicative lifespan, was sometimes impaired in cells from patients with increased tendency to develop tumoral lesions and was sometimes reduced in cells from apparently normal individuals. No differences were found in the fibrin retractile activity of cells from normal embryos or adults. The retractile activity dropped sharply towards the end of the lifespan of some cell lines which could be adequately followed. |
