Summary:A procedure for isolation and purification of prothrombin, applicable to both the bovine and canine species is described. The preparations are of good yield and of very high specific activity, viz. 2400–3000 Iowa units per mg of protein and show a single symmetric component in both the analytical ultracentrifuge and by moving-boundary electrophoresis. Since preparations of such high specific activity are not activated optimally BaCO 3-adsorbed serum, it is necessary to modify the two-stage prothrombin assay method to provide optimal activator activity.
