Summary:1. 1. Hyaluronic acid was isolated from pooled normal and rheumatoid human synovial fluid by a procedure involving fractionation of DEAE-Sephadex, ultra-filtration through sintered glass and fractionation on agarose (2%). 2. 2. Rheumatoid hyaluronic acid formed a gel during ultrafiltration, indicating a specific interaction between the hyaluronic acid and protein. Immunological studies showed that the protein involved was similar to, or identical with, human IgG. Normal hyaluronic acid was not retained by the sintered glass filter. 3. 3. Preparations of purified rheumatoid hyaluronic acid contained 3.5–7.5% protein, and the preparation of normal hyaluronic acid contained 4.6% protein. The amino acid composition of the protein moieties of the rheumatoid and normal hyaluronic acid preparations did not differ significantly.
