Abstract | Short interspersed elements (SINEs), ubiquitous components of eukaryote genomes, can provide primers to PCR amplify DNA segments flanked by these sequences. Species (or family) specificity of SINEs has turned out to be valuable for isolation of species-specific sequences from complex and limited DNA sources by inter-SINE PCR amplification. A high frequency of bovine C-A SINE-elements is known to possess dinucleotide microsatellite tails, suggesting their enrichment among sequences amplified by inter-SINE PCR. In the present paper we describe a SINE-targeting method utilizing this association in microsatellite marker isolation from specific bovine chromosomes. This method may serve as a tool to increase marker density in regions of interest, such as sites of putative QTL, or gaps in the available bovine linkage maps. |
