Gas chromatography-mass spectrometry methods for analysis of 2- and 3-hydroxylated fatty acids: Application for endotoxin measurement

Summary:Gas chromatography-mass spectrometry (GC-MS) methods for analysis of 2- and 3-hydroxy fatty acids were evaluated. These acids were analysed as methyl-trimethylsilyl or methyl-pentafluorobenzoyl derivatives in the electron impact and chemical ionization-negative ion (NICI)_modes, respectively. NICI spectra were strongly influenced by the temperature of the ion source. Both 2- and 3-hydroxylated acids with 10–18 carbon atoms were detected in organic dust collected from a ventilation duct; differential ion monitoring was used for their separation. The described GC-MS methods can be applied for determination of air-borne endotoxins.
