Summary:A method is described for the isolation, identification and frequency evaluation of bacteria inside primary enamel lesions (PEL) of freshly extracted human molars. PEL were localized by transillumination. Molar surfaces were sterilized by ultraviolet light. The PEL and the sound enamel controls were aseptically excavated, cultivated in beef-heart infusion medium and the resulting bacteria were identified by biochemical and gas-chromatographic methods. One hundred and four PEL were studied. Four bacterial genera and eight species were found. The genus Streptococcus displayed the highest frequency, being present in 56.2% of PEL. Actinomyces naeslundii was the most frequent species being present in 25% of PEL followed by S. sanguis 21.9% and S. mitis 18.7%.
