Summary: | The reaction mechanism of erythrocyte phosphofructokinase (PFK) was investigated by the initial velocity and the product inhibition. Intersecting lines obtained with initial velocity studies are consistent with a sequential mechanism and the formation of ternary complex as an intermediate. The product inhibition studies support an ordered Bi Bi mechanism in which fructose 6 phosphate (F6P) is the first substrate binding and adenosine diphosphate (ADP) is dissociated from the enzyme before fructose-1,6-P
2 (FDP).
Le mécanisme de réaction de la phosphofructokinase érythrocytaire (PFK) est étudié par la vitesse initiale en fonction des différentes concentrations de substrats (ATP et F6P) et par l'inhibition par les produits de la réaction (ADP et FDP). Les droites convergentes obtenues par l'étude de la vitesse initiale traduisent un mécanisme séquentiel et la formation d'un complexe ternaire intermédiaire. Les résultats de l'inhibition par les produits de la réaction concordent avec un mécanisme ordonné dans lequel le fructose 6 phosphate (F6P) est le premier substrat à se fixer et l'adénosine diphosphate (ADP) se dissocie de l'enzyme avant le fructose 1–6 diphosphate (FDP). |
