Summary:During the process development of perphenazine, two unknown impurities were found by high‐performance liquid chromatography (HPLC) analysis. These two impurities were further identified, isolated, and characterized form the mother liquor. These two impurities were found novel in the process of perphenazine. Also the reaction conditions were controlled for commercial manufacturing of perphenazine with overall good yield and minimal side reactions with control of these two novel impurities. Developed the efficient and cost‐effective process for the manufacturing of antiemetic drug perphenazine with high purity by removing these two impurities from perphenazine and successfully utilized the process for commercial production of perphenazine in our organization.
