Summary:Affinity matrices, obtained by coupling the ligand phenylacetic acid to Indion 48-R (a macroporous cross linked polystyrene anion exchange resin) through varying spacers, were used to purify penicillin acylase (PA). Homogenous enzyme preparations could be obtained in a single step with over 85% recovery. The effect of spacer variations on the adsorption and elution behaviour of PA is discussed.
