Modified polynucleotides.III. De novo synthesis and hydrolysis of poly d(A-r5U) copolymers

Kinetics of DNA polymerase I enzyme catalyzed de novo synthesis and hydrolysis of poly d(A-r5U) copolymers, where r=H, methyl, ethyl, n-propyl, n-butyl and n-pentyl groups, was studied. Summary:Kinetics of DNA polymerase I enzyme catalyzed de novo synthesis and hydrolysis of poly d(A-r5U) copolymers, where r=H, methyl, ethyl, n-propyl, n-butyl and n-pentyl groups, was studied. Relative rates of polymerization were different from the corresponding rates measured with poly d(A-T) template-primer, which may be the effect of the modified templates. The rate of degradation of the polymers by the exonuclease of DNA polymerase I enzyme depends on the substitution of polynucleotides: with the increase of chain length of r in the polymer, the rate of hydrolysis decreases.
