Summary:In previous studies, we reported an enhancement of ^^14 C-catecholamine synthesis in Na free medium due to the fall in intracellular pH in cultured bovine adrenal medullary cells 「Jap.J.Pharmacol.39,Suppl.375P」. In this study, we investigated the effect of protonophore, FCCP and CCCP on the synthesis of catecholamines and intracellular pH. FCCP and CCCP augmented the synthesis of ^^14 C-catecholamines from ^^14 C-tyrosine and decreased the intracellular pH. The stimulatory effect of FCCP was observed even in Ca free medium. Dibutyryl cAMP increased the synthesis of ^^14 C-catecholamines irrespective of the existence of FCCP. Since the optimal pH for tyrosine hydroxylase activity was observed to be 5.5-6.0, the stimulatory effect of FCCP and CCCP on ^^14 C-catecholamine synthesis may also be due to the fall of intracellular pH as in Na free medium. FCCP: carbonyl cyanide p-trifluoromethoxyphenylhydrazone, CCCP: carbonyl cyanide m-chlorophenylhydrazone.
