Summary:In the present investigation, two new ternary complexes [Cu(II)(dbp)(dmdp)] (1) and [Cu(II)(dbhp)(dmdp)] (2) have been synthesized, characterized, and screened for their biological activities. The complexes were characterized by employing standard spectroscopic techniques. Both the complexes assume square–pyramidal geometry where Schiff bases (dbp and dbhp) and secondary ligand (dmdp) act as ONO and NN donors, respectively. DNA binding studies indicated their high binding propensity in an intercalative mode. Nuclease activity revealed their DNA cleavage ability in presence of an oxidant, hydrogen peroxide. In silico docking studies revealed higher binding affinity for 2 as compared to 1 in the active site pocket of human DNA topo IIβ. In vitro anti-proliferative activity by MTT assay against selected cancer cell lines demonstrated considerable inhibitory efficiency for both the complexes. Anti-bacterial studies revealed higher activity against gram positive bacterial strains. However, the complexes were found to be inactive against gram negative bacterial strain, Escherichia coli.
