Summary: | The crowns of erupting and erupted molars of rats were labelled by injecting the animals with
32P orthophosphate solution. The teeth were extracted, cleaned and their pulp cavities sealed. They were then incubated with streptococci which were caries-inducing in rats in systems containing rat saliva, buffer solutions having two initial pH values or human saliva, and with or without sucrose. In the non-fermenting control systems (i.e. without sucrose), very little of
32P of tooth origin was found in the cells while the majority was in the fluid phase. The presence of sucrose resulted in a pH decrease in the system and in an increased release of phosphorus from the teeth. The pH drop from above 8.0 to slightly acid values was accompanied by an increased uptake of phosphorus, including the majority of tooth origin, by the cells. The total phosphorus in the fluid phase was strongly reduced. The pH drop to 5.3 or lower was accompanied by a greater liberation of tooth phosphorus but the uptake by the cells was less and the majority of
32P was found in the extracellular fluid. The specific activity of
32P in the fermenting cells (cpm/μg total P) was increased 2–31 times above the value of the non-fermenting control cells but it still remained always appreciably below the specific activity of the respective fluid phase.

32P. Les dents sont extraites, nettoyées et leurs pulpes sont obturées. Elles sont ensuite incubées avec des streptocoques, qui induisent des caries chez les rats dans des systèmes contenant de la salive de rat, des solutions tampons ayant deux valeurs initiales de pH ou de la salive humaine, en présence ou en l'absence de saccharose. La chute du pH à 5,3 ou plus bas s'accompagne d'une plus grande libération de phosphore dentaire, mais la fixation au niveau des bactéries est diminuée et la majorité du
32P se retrouve dans le liquide extracellulaire. L'activité spécifique du
32P au niveau des bactéries (cpm/μg de P total) augmente de 2–31 fois au dessus de la valeur des bactéries témoins non fermentantes; mais l'activité reste toujours nettement au dessous de l'activité spécifique de la phase liquide respective.
Die Kronen von durchgebrochenen und durchbrechenden Rattenmolaren wurden durch Injektion von
32P-Orthophosphat markiert. Die Zähne wurden extrahiert, gesäubert und deren Pulpahohlräume gefüllt. Danach wurden sie mit Streptokokken, die bei Ratten Karies verursachen, in Systemen inkubiert, die Rattenspeichel, Pufferlösungen mit zwei initialen pH-Werten, oder menschlichen Speichel mit und ohne Rohrzucker enthielten. In den nicht fermentierenden Kontrollsytemen (ohne Rohrzucker) wurde sehr wenig
32P vom Zahn herstammend in den Zellen gefunden; das meiste fand sich in der flüssigen Phase. Die Anwesenheit von Rohrzucker führte zu einem pH-Abfall im System und zu einer erhöhten Freisetzung von Phosphor aus den Zähnen. Der pH-Abfall von über 8,0 zu leicht sauren Werten war von einer gesteigerten Phosphoraufnahme begleitet, wobei der gröβere Teil des von den Zähnen herstammenden Phosphors von den Zellen aufgenommen wurde. Der Gesamtphosphor der flüssigen Phase war stark reduziert. Der pH-Abfall nach 5,3 oder niedriger wurde von einer gröβeren Freisetzung von Zahnphosphor begleitet, doch war die Aufnahme durch die Zähne vermindert und der Groβteil von
32P wurde in der extrazellulären Flüssigkeit gefunden. Die spezifische Aktivität von
32P in den fermentierenden Zellen (cpm/μg Gesamt-P) war 2-31 mal über den Wert der nicht fermentierenden Kontrollzellen gesteigert, jedoch blieb sie stets noch deutlich unter der spezifischen Aktivität der entsprechenden flüssigen Phase. |
