Summary:Antisolvent crystallization is a significant unit operation in the pharmaceutical industry, especially in drug crystal property optimization. This paper firstly highlights the applications of antisolvent crystallization in crystal engineering. Antisolvent crystallization can effectively control the product properties, such as polymorphism, morphology, and particle size optimization, to improve drug therapeutic effect. This paper also includes the process intensification pathways of antisolvent crystallization in the pharmaceutical field. Process parameter optimization and device and structure modification are considered as feasible approaches. Then, coupled techniques with antisolvent crystallization, including ultrasound and membrane intensification, are also summarized. Finally, the models and applications of continuous antisolvent crystallization are introduced to better design and optimize the processes. Also, critical concerns and opportunities for further research and perspectives are discussed.
