Summary: | Swine X and Y boaring spermatozoa from 9 boars of Duroc, Large Yorkshire and Berkshire, were separated by Percoll density gradient centrifugation. A stock solution of Percoll was diluted with 0.15M-NaCl of the prescribed amount into 40-80% (7 layers). The spermatozoa were washed and loaded onto discontinuous gradients, and centrifugated at 1500rpm for 15 min at room temperature. The upper and bottom layer fractions only were separately collected and stored at 18 degrees C. The concentration and motility of the spermatozoa were examined, but no influence was observed. Twenty two sows and 12 sows were separately inseminated with the spermatozoa of the upper and bottom layer fractions, respectively. The number of offspring was the same as under normal circumstances and the sex ratio was not altered. Thus, the present procedure showed to be ineffective |
