Summary:(8)-Gingerol, a component of ginger, produced a long-lasting contractile response at concentration of 10^-5 M in isolated ileum and trachea from guinea-pig. Contractile response was absolutely dependent on the presence of extracellular Ca^2+ , and was resistant to tetrodotoxin and indomethacin. In ileal longitudinal smooth muscle preparation, (8)-gingerol-induced contraction was completely inhibited in the presence of verapamil or valinomycin. In tracheal preparation, (8)-gingerol-induced contraction was markedly attenuated in the presence of verapamil or valinomycin, and was completely inhibited by supplementation with amiloride. These results suggest that (8)-gingerol mainly acted on Ca^2+ channels of isolated guinea-pig ileal and tracheal smooth muscle membranes. The action may be closely associated with the Ca^2+ influx through voltage-dependent Ca^2+ channels caused by depolarization of smooth muscle membrane.
