Summary:•Dobinin V is the first example of 7,8-seco-11,12,13-trinoreudesmane sesquiterpenoid in nature.•A possible biosynthetic pathway of dobinins V−Z, and AA is proposed.•Lipid-lowering activity of eudesmane sesquiterpenoids was first discovered.•Dobinin AA showed significant in vitro lipid-lowering activity.•Dobinins X and Y showed moderate in vitro antimalarial activities. Six undescribed noreudesmane sesquiterpenoids (dobinins V-Z, and AA; 1–6) were isolated from Dobinea delavayi, including 1 as the first example of 7,8-seco-11,12,13-trinoreudesmane sesquiterpenoids with a unique skeleton fusing by a cyclohexane and a six-membered lactone. NMR and HR-ESI-MS spectroscopies were used to identify the structures of dobinins V-Z, and AA, and single-crystal X-ray diffraction experiment was conducted to verify the structure of dobinin V. A biosynthetic pathway of dobinins V-Z, and AA was suggested based on the structure. The isolated compounds were tested in vitro for lipid-lowering activity and for antimalarial activity against Plasmodium falciparum 3D7. Eudesmane sesquiterpenoids were discovered to have lipid-lowering activity for the first time. [Display omitted]
