Summary:We have designed and synthesized a novel fluorescent oligonucleotide to analyze functions of RNAs. This probe worked as a green fluorophore only when making a stable doublestrand with DNA or RNA. The fluorescence enhancement is rapid, large and target specific. Therefore, this probe has an ideal photophysical property as a fluorescent probe. We applied a fluorescent oligonucleotide to living cells to confirm the condition for introducing the probe. This novel probe and analyzing method will be a very useful tool for understanding RNA functions.
