Summary:Objectives To investigate the effects of intensive atorvastatin on the expression of PDCD4 in CD4 + T lymphocytes from unstable angina patients during perioperative period of PCI. Methods A total of 66 unstable angina patients were enrolled. Unstable angina patients were randomized to pretreatment with intensive-dose atorvastatin (80mg/d, n = 33) or moderate-dose atorvastatin (20mg/d, n = 33). Circulating CD4 + T cells were obtained by magnetic cell sorting system (MACS) at baseline and post- PCI 18-24h. At the gene level, we used real-time fluorescent quantitative polymerase chain reaction (RFQ-PCR) to measure the levels of PDCD4 in CD4 + T lymphocyte. At the protein level, the expression of PDCD4 was detected with Western blot analysis. IL-10 and TNF-α were quantified by enzyme-linked immunosorbent assay (ELISA). Results Compared with intensive-dose group before PCI, the expression of PDCD4 mRNA and protein levels dramatically decreased in intensive-dose group after PCI(P<0.05). 2) Compared with intensive-dose group and moderate-dose group before PCI, the serum level of TNF-α was increased in both groups after PCI. However, level of TNF-α in moderate-dose group was higher than intensive-dose group after PCI(P<0.05). 3) Compared with intensive-dose group and moderate-dose group before PCI, the serum level of IL-10 was increased in both groups after PCI. However, level of IL-10 in intensive-dose group was higher than moderate-dose group after PCI(P<0.05). Conclusions Intensive-dose atorvastatin pretreatment reduced post-PCI myocardial inflammation through inhibiting the expression of PDCD4 in CD4+ T lymphocytes.
