Abstract | S566.1; 类钙调神经素B亚基蛋白(calcineurin B-like proteins,CBLs)是解码钙信号的一类重要蛋白,在植物遭受非生物胁迫过程中发挥重要调节作用.本研究以甘蔗(Saccharum officinarum)'新台糖22号'为实验材料,基于基因转录结果,采用反转录PCR(reverse transcription-PCR,RT-PCR)从低钾胁迫甘蔗根系中克隆得到SsCBL3基因(GenBank No.MN850491).该基因包含1个长度为678 bp的开放阅读框,预测可编码225个氨基酸.对SsCBL3蛋白的序列进行生物信息学分析,显示蛋白分子量为25.76747 kD,理论等电点(isoelectric point,PI)为4.82,预测其属于酸性蛋白.分析SsCBL3的蛋白结构,结果显示,其含有3个的EF-hand(elongation factor hand)结构域和1个与类钙调磷酸酶B互作蛋白激酶(CBL-interacting protein kinase,CIPK)互作的FPSF位点.系统进化树分析结果显示,甘蔗SsCBL3与禾本科作物的亲缘关系较近,与玉米(Zea mays)的亲缘关系最近.实时荧光定量PCR检测结果发现,SsCBL3基因在低氮、低磷、低钾、干旱、盐和脱落酸(abscisic acid,ABA)胁迫过程中的表达均发生变化,并且胁迫时间的不同也会造成基因表达差异.其中,盐和干旱胁迫能够诱导SsCBL3基因强烈表达.qRT-PCR结果说明SsCBL3基因可能在甘蔗遭受逆境胁迫下发挥重要的调控作用.本研究为培育抗逆甘蔗品种提供理论基础. |
