苹果和梨远缘杂种后代的鉴定
S662; 种间远源杂交是创制果树新种质的方法之一,但是如何检测真正的种间杂交种一直缺乏高效、可靠的技术手段.本研究在基因组学研究的基础上,通过苹果(Malus)和梨(Pyrus)的全基因组序列比较分析,基于两者间的插入缺失位点,设计InDel特异性分子标记,结合PCR检测,筛选出6对特异性高、重复性好且扩增条带清晰的特异性分子标记.其中,3对标记引物来自梨基因组,3对标记引物来自苹果基因组,可有效检测两者间差异.利用上述6对特异分子标记对3组苹果和梨属间杂交后代的311株个体进行鉴定,结果表明7株后代可同时扩增出苹果和梨特异条带,确定为属间杂交种,占总杂交后代的2.3%.其中,'红...
