C34 MECHANISMS OF HOST DEFENSE IN BACTERIAL AND FUNGAL INFECTIONS: Targeted Blockade Of The Programmed Cell Death-1 Receptor Improves Fungal Clearance In A Murine Model Of Persistent Cryptococcal Lung Infection

Summary: | Rationale: The Programmed cell death-1 (PD-1) signaling pathway modulates adaptive effector immune responses within tumors and persistently-infected tissue microenvironments. At 35 and 49 dpi, mice were euthanized and evaluated for the following immune parameters (using CFU assay, 12-parameter flow cytometric analysis, and regression analysis): 1) lung, brain, and spleen fungal burden, 2) the abundance of numerous lung myeloid and lymphoid subsets, 3) macrophage and dendritic cell activation (assessed by expression of MHC Class II, CD80, iNOS, and arginase), and 4) T cell polarization and activation (assessed by expression of ICOS, OX40 and transcription factors). |
