Abstract Resistance to Pseudomonas syringae pv. tomato (Pst) in tomato is conferred by a single dominant locus, Pto (Pitblado et al., 1984 Martin et al., 1993a). The Pto locus interacts in a gene-for-gene fashion with an avirulence gene, avrPto, that has been isolated from Pst (Ronald et al., 1992). As with most disease resistance loci, nothing was known of the products that may be encoded at the Pto locus and this precluded standard methods of gene isolation. In order to isolate Pto, a multi-step "map-based cloning" strategy was used.
