Abstract Pollen and anther cultures are most popular methods for obtaining haploid plants and later on double haploid (DH). DH lines are homozygous in all loci, therefore it is possible to do a quick true evaluation of the breeding initial material and, subsequently, reduced time period of the breeding process. In the Laboratory of Plant Genetics of the Institute of Biology, University of Latvia, anther pollen and embryo culture methods were used for obtaining of DH lines from barley hybrids produced in Stende and Priekuli Plant Breeding Stations. Powdery mildew is one of the widespread barley diseases in Latvia. Genetic resistance is an important alternative to fungicides. Latvian barley varieties have not good resistance genes against the corresponding pathogen. Hordeum vulgare ssp. spontaneum can be used as an additional source of resistance to powdery mildew for Latvian barley breeding. To accelerate introduction of new resistance genes in the local material the use of DH technique is very essential. In this article we are presenting the first results of testing three hybrid combinations of 'Ruja' x Hordeum vulgare ssp. spontaneum, resistant to powdery mildew, for calli and embryo development in anther cultures. It was found that three investigated hybrids have different ability to callusogenesis and embryogenesis and DH plant regeneration. Several DH plants were obtained from one of the hybrid (Sp2).
