Abstract | In order for callus production and regeneration in spinach effects of genotype, different plant hormone compositions and phisical conditions were studied. Different levels of IAA as well as low levels of GA3 showed the highest amount of callus induction, while application of high levels of IAA and GA3 produced the lowest amount of callus. Also low level of IAA in the induction medium significantly (α=5%) rised the regeneration rate. Culture of hypocotyle segments first on medium containing 15 mg/l IAA and 3/4 mg/l GA3 subsequently transfered to regeneration medium (2 mg/l IAA and 3/4 mg/l GA3) showed the highest regeneration rate. Culture of root, hypocotyle, cotyledon and leaf tissues on different compositions, NAA, 2,4-D with TDZ and or BAP produced only callus. Different levels of MS and GA3 showed significant difference effects on regeneration. Decreasing MS concentration to a half amount showed embryogenesis. For shoot tip growth IAA and BAP levels and their interactions was significant at 1% level. The highest regeneration amount was obtained at 0.02 mg/l BAP. For cytogentic studies a procedure for chromosome slide preparation was optimized. |
