Abstract In order to improvement and introduction of a new potato cultivar, five potato cultivars Agria, Kayser, Borene, Savalan and Kenebek were crossed in reciprocal method. Concurrent to crossing, two cultivars Kenebek and Banba were radiated with different dosages of gamma ray. For crossing, tubers of different cultivars were put on separate bricklayer and covered with suitable sowing bed in 2006 and springtime. Irrigation was done using tape method and plants weekly sprinkled with complete fertilizer. For changing sink and source relations, plants before flowering were sprinkled by 500 ppm gibberellic acid and tubers were harvested every two weeks after tuberization. Pollination was done after sterilizing and fruits (berries) separately harvested 45 days after pollination. Assessment of segregating population, single plant and preliminary study were carried out by 2011. Finally, 69 superior clones were selected to use in advance study. Out of 69 clones, 41 obtained from __Kayzer x __Savalan, 3 from __Borene x __Savalan, 3 from __Savalan x __Kayzer, 4 from __Kayzer x __Kenebek, 6 from __Savalan x __Agria and 12 from __Kayzer x __Agria. For mutation, the healthy and free of virus buds of Kenebek and Banba were cultured in modified MS media under sterilized condition. The healthy plants were recognized using ELISA test and transferred to gamma cell after propagation and exposed to 20 and 25 gray of gamma ray. After 3 genaration, 1500 microtubers were obtained from each cultivar. The micro tubers were planted in plastic pots under greenhouse condition and 768 clones of Banba and 1091 clones of Kenebek were selected to check under field condition.
