Abstract The invention provides surface plasmon resonance-mass spectrometry for the rapid, sensitive and accurate investigation of molecular interactions coupled with the identification and quantification of the same. Methods of the invention include capturing an analyte present within a sample by an interactive surface layer on a real-time interaction analysis sensor, analyzing the analyte by surface plasmon resonance while the analyte is captured by the interactive surface layer, and identifying the captured analyte by desorbing/ionizing the analyte from the interactive surface layer while under vacuum within a mass spectrometer. Devices of the invention include a transparent material, a conductive material capable of supporting surface plasmon resonance affixed to the transparent material, an interactive surface affixed to the conductive material, and a means for exposing the interactive surface to the interior of a mass spectrometer without breaking the vacuum therein. L'invention concerne la spectrométrie de masse par résonance de plasmons de surface. Ce type de spectrométrie permet d'étudier avec rapidité, sensibilité et précision les interactions moléculaires tout en les identifiant et en les quantifiant. Les procédés selon l'invention consistent à capturer un analyte présent dans un échantillon par une couche de surface interactive sur un capteur d'analyse d'interactions en temps réel, à analyser l'analyte par résonance de plasmons de surface tandis que l'analyte est capturé par la couche de surface interactive, et à identifier l'analyte capturé en désorbant/ionisant l'analyte à partir de la couche de surface interactive, tout en le maintenant sous vide dans un spectromètre de masse. Les dispositifs selon l'invention comprennent un matériau transparent, un matériau conducteur permettant la résonance de plasmons de surface, fixée au matériau transparent, une surface interactive fixée au matériau conducteur, et un moyen pour exposer la surface interactive à l'intérieur d'un spectromètre de masse sans rompre le vide dans ce dernier.
