Abstract | The present invention relates to the field of nucleic acid production, in particular in vitro RNA transcription. More specifically, the present invention relates to a method wherein physicochemical properties of the reaction mixture are monitored in order to timely stop the reaction and/or to add fresh reagents to allow the reaction to continue under optimal conditions. In particular, conductivity, but also pH and/or optical density is determined at at least two different time points, or continuously during the in vitro transcription reaction.
La présente invention concerne le domaine de la production d'acides nucléiques, en particulier la transcription d'ARN in vitro. Plus particulièrement, la présente invention concerne un procédé permettant de surveiller les propriétés physicochimiques du mélange réactionnel afin d'arrêter la réaction en temps opportun et/ou d'ajouter des réactifs frais pour permettre à la réaction de se poursuivre dans des conditions optimales. En particulier, la conductivité, mais aussi le pH et/ou la densité optique sont déterminés à au moins deux moments différents, ou en continu pendant la réaction de transcription in vitro. |
