Abstract The present invention provides: a method for correcting a microalgae gene without DNA introduction by using a RNA-guided engineered nuclease (RGEN) RNP complex; and a microalgae variant with a corrected gene. The gene correction method according to the present invention has an advantage in that a desired gene can be selectively and accurately corrected without DNA introduction. In addition, the present invention is directed to novel algae with an improved pigment producing ability, and the use of a mutant strain of the present invention can produce carotenoid-based pigments, specifically, xanthophylls, through the consumption of less energy, thus leading to an efficient production of pigments at the industrial level. In addition, the present invention can be applied as a raw material for food, health functional food, and medicine containing pigments. Especially, a procedure of manufacturing the mutant strain is not assumed to be regulated by GMO legislation since DNA fragments are not likely to be inserted into a target nucleotide sequence or a non-target nucleotide sequence. Therefore, it is expected that the present invention will have a great economic effect in an industrial aspect in which lutein and zeaxanthin are produced using microalgae. La présente invention concerne : un procédé de correction d'un gène de micro-algues sans introduction d'ADN à l'aide d'un complexe de nucléase modifiée guidée par ARN (RGEN)-RNP; et un variant de micro-algues présentant un gène corrigé. Le procédé de correction de gène selon la présente invention présente un avantage en ce qu'un gène souhaité peut être corrigé de manière sélective et précise sans introduction d'ADN. De plus, la présente invention concerne de nouvelles algues présentant une aptitude de production de pigment améliorée et l'utilisation d'une souche mutante de la présente invention peut produire des pigments à base de caroténoïdes, plus particulièrement, des xanthophylles, par la consommation de moins d'énergie, ce qui conduit à une production efficace de pigments au niveau industriel. De plus, la présente invention peut être appliquée en tant que matière première pour des aliments, des aliments fonctionnels de santé et des médicaments contenant des pigments. En particulier, un procédé de fabrication de la souche mutante n'est pas supposé être régulé par la législation sur les OGM, puisqu'il n'y a pas de probabilité d'insertion de fragments d'ADN dans une séquence nucléotidique cible ou dans une séquence nucléotidique non cible. Par conséquent, il est attendu que la présente invention aura un grand effet économique dans un aspect industriel dans lequel la lutéine et la zéaxanthine sont produites à l'aide de micro-algues. 본 발명은 RGNE (RNA-guided engineered nuclease) RNP의 복합체를 이용하여 DNA 도입 없이 미세조류의 유전자를 교정하는 방법, 및 유전자가 교정된 미세조류 변이체를 제공한다. 본 발명에 따른 유전자 교정 방법은 DNA 도입 없이 원하는 유전자를 정확하게 선택적으로 교정할 수 있다는 이점을 갖는다. 또한, 본 발명은 색소 향상된 색소 생산능을 갖는 새로운 조류에 관한 것으로, 본 발명의 변이주를 이용하면 적은 에너지를 소모하여, 카로티노이드계 색소, 구체적으로 잔토필을 생산하는 것이 가능하므로 산업 수준에서 효율적으로 색소를 생산할 수 있다. 또한, 색소가 포함되는 식품, 건강기능식품 및 의약품의 원료로 적용이 가능하다. 특히, 상기 변이주가 제작되는 과정은 DNA 조각이 표적 염기서열 또는 표적외 염기서열 내에 삽입될 가능성이 없기 때문에 GMO로 규제되지 않을 것으로 예상되므로 미세조류를 이용하여 루테인과 지아잔틴을 생산하는 산업적인 측면에서 큰 경제 효과를 낼 수 있을 것으로 기대된다.
