Abstract The present invention concerns a new analytical method for qualitative and quantitative detection of short nucleic acid sequences, preferably a DNA oligonucleotide or a modified DNA oligonucleotide such as antisense oligonucleotides or fragmented nucleic acid sequences of about 8-50 nucleotides in length, from especially biological sample material. The invention relates to the introduction of modified nucleic acids into an oligonucleotide probe that hybridizes to the target sequence such that amplification and quantitation of the short nucleic acid sequence is enabled and sensitivity and specificity of the reaction is increased. The invention also embraces test kits for performing nucleic acid amplification to detect and quantitate short nucleic acid sequences and processes for preparing such and the use of new analytical methods. La présente invention concerne un nouveau procédé analytique de détection qualitative et quantitative de courtes séquences d'acides nucléiques, de préférence d'un oligonucléotide d'ADN ou d'un oligonucléotide d'ADN modifié tel que des oligonucléotides antisens ou des séquences d'acides nucléiques fragmentées présentant une longueur approximative de 8 à 50 nucléotides, à partir d'un échantillon, en particulier biologique. L'invention concerne notamment l'introduction d'acides nucléiques modifiés dans une sonde oligonucléotidique qui s'hybride avec la séquence cible de façon à permettre l'amplification et l'analyse quantitative de la courte séquence d'acides nucléiques et accroître la sensibilité et la spécificité de la réaction. L'invention concerne en outre des trousses d'analyse pour mettre en oeuvre l'amplification d'acides nucléiques afin de détecter et d'analyser quantitativement de courtes séquences d'acides nucléiques, des processus de préparation associés, ainsi que l'utilisation de nouveaux procédés analytiques.
