Abstract The present invention provides a method for amplifying at least one target gene having a GC content of 60% or greater in a reaction solution, wherein: the method includes a step for amplifying a base sequence specific to a target gene using a primer set composed of a first primer and a second primer for amplifying the target gene; one of the primers in the primer set is added to the reaction solution in a higher molar ratio than the other primer; and neither of the primers is a hot-start primer that includes an inactivating chemical modification restored by the action of an activating enzyme.
