Provided herein is an immunocytokine comprising (i) an antigen binding protein (ABP) specific to a target protein; (ii) an IL-21 domain; and (iii) an IL-21Rα mutein, wherein the IL-21Rα mutein has a reduced binding affinity to the IL-21 domain compared to a wild-type IL-21Rα. Further provided includes a method of using the immunocytokine to selectively activate an IL-21Rα on a target cell, thereby enhance immune response or treat cancer. En el presente documento, se proporciona una inmunocitocina que comprende (I) una proteína de unión a antígeno (ABP) específica a una proteína objetivo, (II) un dominio IL-21 y (III) una muteína de IL-21Ra, donde la muteína de IL-21Ra tiene una afinidad de unión reducida al dominio IL-21 en comparación a una IL-21Ra de tipo natural. Se proporciona, además, la inclusión de un método para usar la inmunocitocina para activar, selectivamente, una IL-21Ra en una célula efectora para, de este modo, aumentar la respuesta inmunitaria o tratar el cáncer.
