Summary: | The present specification provides a method for screening an immune checkpoint inhibitor, which comprises the following steps: firstly culturing a biological sample isolated from an individual so that T cells are enriched; treating the enriched T cells with an immune checkpoint inhibitor; mixing the T cells treated with the immune checkpoint inhibitor and cancer cell line in which a gene for an immune checkpoint ligand is knocked out, and secondly culturing the T cells; and measuring the amount of cytokine secretion of the secondly cultured T cells. The method for screening an immune checkpoint inhibitor of the present invention can clearly identify the difference in the amount of cytokine secretion through a modified knockout cancer cell line and T cells, and thus an effect of the immune checkpoint inhibitor can be intuitively evaluated.
본 명세서에서는 개체로부터 분리된 생물학적 시료를 T세포가 농축(enrichment)되도록 제 1 배양하는 단계, 농축된 상기 T세포에 면역 관문 저해제(immune checkpoint inhibitors)를 처리하는 단계, 면역 관문 저해제가 처리된 상기 T세포와 면역관문 리간드(immune checkpoint ligand)에 대한 유전자가 녹아웃(knock-out)된 암 세포주를 혼합하여 제 2 배양하는 단계, 및 제 2 배양된 상기 T세포의 싸이토카인 분비량을 측정하는 단계를 포함하는, 면역 관문 저해제의 스크리닝 방법을 제공한다. |
