Summary:The present invention relates to a method for measuring damage to skin DNA by ultraviolet radiation (UV) rays from DNA extracted from the stratum corneum of the skin, and a use thereof. The method for measuring damage to skin DNA using the stratum corneum of the skin according to the present invention comprises the steps of: collecting samples of the stratum corneum from the skin; immersing the collected samples of the stratum corneum in a dissociation buffer; warming the dissociation buffer; treating with antibody the DNA, which is present in the keratinocyte separation solution obtained by centrifugation of the dissociation buffer; and detecting a complex of a DNA damage marker and the antibody. The samples are easily obtained in a non-invasive way and damage to skin DNA is measured, so that it can be used for analyzing the degree of skin sensitivity due to UV rays. 본 발명은 피부 각질층에서 추출한 DNA로부터 자외선으로 인한 피부 DNA의 손상 여부를 측정하는 방법 및 이의 용도에 관한 것으로서, 피부로부터 각질층을 채취하는 단계, 채취된 각질층을 해리 완충용액에 침지하는 단계, 해리 완충용액을 가온하는 단계, 이를 원심분리하여 수득한 각질세포 분리액 내 존재하는 DNA에 항체 처리하는 단계, DNA 손상 마커와 항체의 복합체를 검출하는 단계를 포함하며, 비침습적인 방법으로 간편하게 시료를 획득하여, DNA 손상 여부를 측정하고, 자외선에 의한 피부 민감도 분석에 활용할 수 있는 이점을 제공한다.
